چکیده

Over the years, fluorescence microscopy has evolved and become a necessary element of life science studies. Microscopy elucidated biological processes in live cells organisms, also enabled tracking biomolecules real time. Development highly sensitive photodetectors light sources, addition to evolution various illumination methods fluorophores, helped acquire single-molecule sensitivity, enabling imaging detection. Low-light used are classified into two categories: point wide-field photodetectors. Although photodetectors, notably photomultiplier tubes (PMTs), have been commonly laser scanning (LSM) with confocal setup, such as electron-multiplying charge-coupled devices (EMCCDs) scientific complementary metal-oxide-semiconductor (sCMOS) cameras imaging. This review focuses on former low-light presents their applications recent progress. These include conventional PMTs, single photon avalanche diodes (SPADs), hybrid (HPDs), newly emerging silicon photomultipliers (SiPMs) (also known multi-pixel counters (MPPCs)) superconducting nanowire detectors (SSPDs). In particular, this shows distinctive features HPD application

Two-photon Fluorescence Light Microscopy

The invention of two-photon fluorescence light microscopy by Denk, Webb and co-workers (Denk et al., 1990) revolutionized three-dimensional (3D) in vivo imaging of cells and tissues. In 1931, the theoretical basis of twophoton excitation was established by Maria GöppertMayer, and this photophysical effect was verified experimentally by Kaiser and Garret in 1963. Two-photon excitation is a fluor...

Scattered light fluorescence microscopy in three dimensions.

Recently, we have proposed a method to image fluorescent structures behind turbid layers at diffraction limited resolution using wave-front shaping and the memory effect. However, this was limited to a raster scanning of the wave-front shaped focus to a two dimensional plane. In applications, it can however be of great importance to be able to scan a three dimensional volume. Here we show that ...

Spatial light modulators in fluorescence microscopy

Spatial light modulators (SLMs) are becoming increasingly important in optical microscopy. These reflective or transmissive devices, based on microdisplays using liquid crystal or micromirror array technology, are used to achieve a variety of useful effects and imaging modes. When placed in an image plane, SLMs can be used to create arbitrary, computer controlled masks. These masks can then be ...

Low-Cost Cryo-Light Microscopy Stage Fabrication for Correlated Light/Electron Microscopy

The coupling of cryo-light microscopy (cryo-LM) and cryo-electron microscopy (cryo-EM) poses a number of advantages for understanding cellular dynamics and ultrastructure. First, cells can be imaged in a near native environment for both techniques. Second, due to the vitrification process, samples are preserved by rapid physical immobilization rather than slow chemical fixation. Third, imaging ...

Breaking the diffraction barrier in fluorescence microscopy at low light intensities by using reversibly photoswitchable proteins.

Fluorescence microscopy is indispensable in many areas of science, but until recently, diffraction has limited the resolution of its lens-based variant. The diffraction barrier has been broken by a saturated depletion of the marker's fluorescent state by stimulated emission, but this approach requires picosecond laser pulses of GW/cm2 intensity. Here, we demonstrate the surpassing of the diffra...
